Testing DMT and Tryptamines: Ehrlich, Hofmann, and Distinguishing N,N-DMT from 5-MeO-DMT

DMT and 5-MeO-DMT look identical but have vastly different dose ranges and effect profiles. Here is the reagent protocol for telling them apart — and why the distinction matters.

Why Distinguishing Tryptamines Matters

N,N-DMT (the classic DMT experience) and 5-MeO-DMT (sometimes called "the God molecule") are both tryptamines, both found in nature, both active when vaporized — and completely different in potency, duration, and effect.

The dose of N,N-DMT that produces a breakthrough experience is 25-50mg. The dose of 5-MeO-DMT that produces a comparable intensity is 5-15mg. Confusing the two — or assuming they are the same — can result in an experience far more intense than intended, with risk of vomiting, loss of bodily control, and psychological distress.

The Reagent Protocol

Ehrlich Reagent (Indole Detection)

Ehrlich reagent reacts with indole rings — the chemical structure common to DMT, 5-MeO-DMT, bufotenine, psilocybin, LSD, and most other tryptamines.

  • All tryptamines: Purple (confirms indole presence)
  • No reaction = not a tryptamine. The substance is something else entirely

Ehrlich cannot distinguish between different tryptamines. It tells you what class you are dealing with — not which specific compound.

Hofmann Reagent (Secondary Tryptamine Screen)

Hofmann is structurally related to Ehrlich and also reacts with indoles, but produces different colors with different tryptamines:

  • N,N-DMT: Blue-purple
  • 5-MeO-DMT: Green → purple (slower, different initial color)
  • Psilocybin/psilocin: Blue-purple
  • LSD: Blue-purple

Marquis Reagent (Differentiating Tryptamines)

  • N,N-DMT: Yellow → brown
  • 5-MeO-DMT: Dark brown (deeper and faster than N,N-DMT)
  • Bufotenine: Brown

Mecke Reagent (Further Differentiation)

  • N,N-DMT: Yellow → brown → dark green
  • 5-MeO-DMT: Dark green → brown (different color sequence)
  • Other tryptamines: Variable

The Testing Protocol

  1. Ehrlich first. Purple = tryptamine. No reaction = not a tryptamine — stop here, this is not DMT.
  2. Marquis second. Yellow/brown = likely N,N-DMT. Dark brown = possibly 5-MeO or bufotenine.
  3. Mecke third. Green → brown progression = consistent with N,N-DMT.
  4. Hofmann (if available): Blue-purple = N,N-DMT. Green-purple = 5-MeO-DMT.

Why This Matters: The Dosage Difference

If you cannot determine whether you have N,N-DMT or 5-MeO-DMT:

  • Assume it is 5-MeO-DMT (the more potent option)
  • Start with 5mg or less (a tiny amount — roughly the size of a match head)
  • Observe effects fully before considering more
  • Understand that 5-MeO-DMT produces a qualitatively different experience — it is more likely to involve ego dissolution at lower doses and more likely to produce somatic effects (body tremors, vocalizations)

DMT Vape Cartridges: The New Variable

DMT vape cartridges are increasingly common and introduce additional variables:

  • Concentration varies enormously — from 1:1 DMT-to-PG to 4:1
  • The solvent (PG/VG) can interfere with reagent testing — dilute before testing
  • Some cartridges contain MAOIs (ayahuasca-style blends) — these alter the duration and intensity significantly
  • Testing a vape cartridge with reagents is messy. Extract a tiny amount of the concentrate if possible and test that directly

Safety Considerations

  • Never vaporize DMT alone. The experience can involve loss of bodily control. Have a sitter.
  • Use a device that cannot break or cause fire if dropped — you may lose motor control during the experience.
  • Do not stand or walk during the experience. Sit or lie down before inhaling.
  • If using MAOI-containing blends (changa, ayahuasca vapes): Research dietary and medication interactions. MAOIs combined with certain foods, medications (SSRIs especially), or other drugs can be fatal.

DMT is one of the most intense psychedelic experiences available. Knowing what you have — and dosing accordingly — is the foundation of a safe experience.
