Testing DMT and Tryptamines: Ehrlich, Hofmann, and the Two-Stage Protocol

DMT, 5-MeO-DMT, and bufotenine all react differently to the same reagents. The two-stage protocol that tells them apart — and why vape cartridges add a new layer of risk.

Same Class, Very Different Substance

The tryptamine chemical family includes compounds with vastly different potency, duration, and safety profiles. DMT (N,N-dimethyltryptamine) produces a short-acting immersive experience measured in minutes. 5-MeO-DMT is substantially more potent per milligram and produces a qualitatively different experience. Bufotenine — also a tryptamine — has its own profile. In an unregulated market, misrepresenting one as another is trivial because they all look like off-white powder or crystal.

Testing is the only way to know which tryptamine is in the bag, and every tryptamine user should also be aware of contamination risk in the newer vape cartridge form factor.

The Two-Stage Protocol

Two independent reactions from two chemically different reagents produces a result you can act on. The same pair of reagents that rules out NBOMe on a tab also confirm DMT in powder — the chemistry is the same; only the reference chart changes.

DMT Vape Cartridges: A Multi-Layered Problem

DMT in vape cartridges is a relatively new product form, and cartridges present an unusually complex testing challenge because there are now three separate things to worry about:

  • The active compound — is this actually DMT or a cheaper tryptamine?
  • The carrier liquid — what is the DMT dissolved in, and is it safe to heat?
  • Potential contaminants — cartridges require several processing steps; cross-contamination has been flagged by some drug checking programs

Testing a cartridge means extracting a tiny amount of the liquid for reagent testing. For full analysis a spectrometry-based drug checking service is the better tool. For home testing, the DMT kit + a fentanyl strip covers the most dangerous failure modes.
